The optimized carbapenem inactivation method for objective and accurate detection of carbapenemase-producing Acinetobacter baumannii

چکیده
The modified carbapenem inactivation method (mCIM) recommended by the Clinical and Laboratory Standards Institute is not applicable for detecting carbapenemases in Acinetobacter baumannii . Four currently reported phenotypic detection methods, namely, Hodge test, mCIM, adjusted simplified (sCIM), did perform well our 90 clinical A. isolates. Thus, minimal inhibitory concentrations (MICs) of carbapenems existence expression carbapenemase-encoding genes were detected to explain results. According E-test, which was more accurate than VITEK 2 system, 80.0 41.1% resistant imipenem (IPM) meropenem (MEM), respectively, 14.4 53.3% exhibited intermediate resistance, respectively. Five β-lactamase found, bla OXA-51-like , TEM OXA-23-like frequently 85 non-susceptible strains. positively correlated with MIC values IPM MEM. Therefore, an improved approach based on designated optimized CIM (oCIM), developed this study detect accurately reproducibly. condition evaluating factors inoculum, incubation broth volume, MEM disk time. Obvious high sensitivity (92.94%) specificity (100.00%) obtained using oCIM, cost-effective reproducible routine laboratory work.

Retrospective and prospective evaluation of the Carbapenem inactivation method for the detection of carbapenemase-producing Enterobacteriaceae
BACKGROUND There is an urgent need for accurate and rapid diagnostic tests to identify carbapenemase producing enterobacteria (CPE). Here, we have evaluated the Carbapenem Inactivation Method (CIM) test to detect CPEs from cultured colonies. METHODS A total of 256 enterobacterial isolates were used to evaluate the performance of the CIM in comparison to Carba NP test and molecular detection u...
متن کاملDetection of carbapenemase-producing Acinetobacter baumannii in a hospital.
Acinetobacter baumannii strains resistant to both imipenem (IPM) and ceftazidime (CAZ) were isolated from 1994 through 1996 at Gunma University Hospital. Nine isolates from different inpatients were examined for carbapenem-hydrolyzing activity and for the carbapemase gene bla(IMP) by the PCR method. All nine isolates were carbapenemase-producing strains that hydrolyzed IPM and that harbored bla...
متن کاملMulticity outbreak of carbapenem-resistant Acinetobacter baumannii isolates producing the carbapenemase OXA-40.
During 2005 we detected a multicity outbreak of infections or colonization due to high-level imipenem-resistant Acinetobacter baumannii (MIC, 64 microg/ml). One hundred isolates from diverse sources were obtained from seven acute-care hospitals and two extended-care facilities; 97% of the isolates belonged to one clone. Susceptibility testing of the first 42 isolates (January to April 2005) rev...
متن کاملOutbreak of infection by carbapenem-resistant Acinetobacter baumannii producing the carbapenemase OXA-58 in Belgium.
Carbapenem-resistant Acinetobacter baumannii isolates were obtained from 17 patients between September 2004 and August 2005 at the Academisch Ziekenhuis Vrije Universiteit Brussel, Brussels, Belgium. These multidrug-resistant isolates, which belonged to a single clone, remained susceptible to colistin and tigecycline only and produced the carbapenem-hydrolyzing oxacillinase OXA-58. This study h...

Background: We evaluated the carbapenem inactivation method (CIM) compared with the modified Hodge test (MHT) for the detection of carbapenemase-producing Gram-negative bacilli. Methods: A total of 61 isolates of carbapenemaseproducing Enterobacteriaceae (CPE: 14 KPC, 7 GES5, 8 NDM-1, 9 VIM-2, 9 IMP-1, and 14 OXA-48-like), 34 isolates of metallo-β-lactamase (MBL)-producing Pseudomonas spp. (14 ...
